ISO 11701:2009 specifies a method for the quantitative determination of phospholipids content by high performance liquid chromatography (HPLC) using a diol column and a light-scattering detector. The method is applicable to crude, oil-containing lecithins, and to oil-free lecithins and lecithin fractions from vegetable fats and oils. The method is not applicable to animal and ruminant lecithins and enzymatically hydrolysed lecithins as the peak separation of lysophosphatidylethanolamine (LPE), lysophosphatidylinositol (LPI) and lysophosphatidic acid (LPA) is insufficient.
Abstract
Overview
ISO 11701:2009, published by the International Organization for Standardization (ISO), specifies a standardized method for the quantitative determination of phospholipids content in lecithins derived from vegetable fats and oils. The method utilizes high performance liquid chromatography (HPLC) equipped with a diol column and a light-scattering detector. It is applicable to various types of lecithins, including crude, oil-containing, oil-free, and fractionated lecithins from vegetable origins. However, it is not suitable for animal, ruminant, or enzymatically hydrolysed lecithins, due to insufficient separation of certain phospholipids.
This international standard facilitates consistent, accurate, and reproducible results in laboratories and quality control settings, underpinning the integrity of food ingredients and supporting compliance with global best practices in the food and oils industry.
Key Topics
-
Phospholipids Analysis
The standard focuses on the measurement of key phospholipids such as N-acyl-phosphatidylethanolamine, phosphatidic acid, phosphatidylethanolamine, phosphatidylcholine, phosphatidylinositol, and lysophosphatidylcholine. -
HPLC Methodology
- Utilizes a diol-silica column for effective separation
- Employs a light-scattering detector to identify and quantify phospholipids
- Follows precise calibration and sample preparation procedures
- Ensures accuracy through use of certified reference materials
-
Scope of Application
- Designed specifically for vegetable-derived lecithins
- Not applicable to animal lecithins or enzymatically altered lecithins
-
Precision and Repeatability
- Includes guidelines to assure test repeatability and reproducibility across laboratories
- Provides standard procedures for calibration and reporting
Applications
ISO 11701:2009 provides crucial guidance for several sectors:
-
Quality Control in Food Manufacturing
Ensures that lecithin ingredients meet required phospholipid content standards, supporting product consistency and regulatory compliance. -
Ingredient Authentication
Facilitates the verification of lecithin source and composition, helping detect adulteration or mislabeling in vegetable oils and fats. -
Research and Product Development
Supports R&D in refining lecithin extraction processes, improving functional or nutritional properties of food ingredients. -
Laboratory Analysis and Accreditation
Laboratories seeking accreditation or optimizing their internal protocols use this method for reliable, cross-comparable data.
