ISO 13212:2014 provides guidance on the design of stability studies for use in gathering information to enable determination of the shelf-life of contact lens care products.
Abstract
Overview - ISO 13212:2014 (contact lens care products shelf-life)
ISO 13212:2014 provides guidance on designing stability studies to determine the shelf-life of contact lens care products. Focused on ophthalmic optics, the standard helps manufacturers gather the chemical, physical, and microbiological data needed to define storage conditions and expiry dates for solutions, suspensions, tablets and other contact lens care formulations. It is a guidance standard (third edition, 2014) and does not cover in-use (post-opening) discard dates.
Key topics and technical requirements
- Objective: Generate reliable data to establish the period during which product performance, safety and acceptability are maintained.
- Study design: Real-time and accelerated/variable-condition studies are recommended. Real-time testing typically targets temperatures between 20 °C and 30 °C with 25 °C ± 2 °C as the mean kinetic testing temperature.
- Analytical methods: Use validated, stability‑indicating methods capable of distinguishing active ingredients from degradation products; test procedures must be fully described.
- Batches and packaging: Normally test three representative finished-product batches in the primary containers intended for marketing; smallest container size (highest surface/volume) is prioritized.
- Environmental conditions: Consider multiple temperatures, humidity ranges (including high humidity up to 75 % ±5 % for solids and low humidity for aqueous systems), light exposure, freeze–thaw cycles, and refrigerated/freezer conditions where relevant.
- Characteristics to monitor: Physical (pH, clarity, viscosity, moisture), chemical (assay of actives, preservatives, antioxidants), microbiological (antimicrobial activity, preservative efficacy, sterility, microbial limits), container–closure interactions, and relevant performance tests when chemical markers are insufficient.
- Data and reporting: Summarize results (tables/graphs), provide initial and time‑point data for each batch, and include discussion, interpretation and justification of proposed shelf-life. A minimum of three months real-time data at 25 °C, supported by accelerated studies, is required; short-term data are not normally adequate to predict shelf-life beyond two years.
Applications - who uses ISO 13212:2014
- Contact lens care product manufacturers (formulation scientists, QA/QC, regulatory affairs)
- Packaging and materials engineers assessing container–closure impact
- Analytical laboratories developing stability-indicating assays
- Regulatory authorities and auditors reviewing shelf-life claims and labeling
- Product development teams planning stability-testing plans and shelf-life extension strategies
