ISO 16603:2004 describes a laboratory test method for measuring the penetration resistance of clothing materials to blood and body fluids. This test method uses a synthetic blood in continuous contact with the material specimen at specified set of conditions using the ISO 13994 test apparatus. This test method is not always effective in testing protective clothing materials having thick, inner liners which readily absorb the synthetic blood.
Abstract
Overview - ISO 16603:2004 (synthetic blood penetration test)
ISO 16603:2004 specifies a laboratory test method to determine the resistance of protective clothing materials to penetration by blood and body fluids using a synthetic blood simulant. The method uses the ISO 13994 penetration test cell and exposes material specimens to synthetic blood under defined time and pressure sequences. Results are reported as pass/fail for visible synthetic blood penetration. The standard is intended for comparative assessment of material barrier performance and includes guidance on specimen selection, conditioning and test procedure.
Key topics and technical requirements
- Synthetic blood properties: surface tension (0.042 ± 0.002 N/m), pH (7.3 ± 0.1), viscosity (2.7 ± 0.3 mPa·s), conductivity (12.0 ± 1.2 mS/cm). A formula is provided in Annex A.
- Test principle: material acts as a partition in a pressurized cell; any visible synthetic blood on the viewing side = failure.
- Apparatus: ISO 13994 penetration test cell (≈60 ml capacity), retaining screen (open area >50 %, specimen deflection ≤ 5.0 mm), air pressure source up to 20.0 kPa, timer, balance (±0.01 g), thickness gauge (±0.02 mm).
- Specimen preparation: cut squares (min. 70 mm, preferred 75 mm); test three specimens per material/location. Condition specimens 24 h at (21 ± 5) °C and (60 ± 10) % RH.
- Sealing and test area: seal specimen edges (adhesive, parafilm, wax) when needed to prevent wicking; leave a central 57 mm square exposed for testing.
- Test pressures and procedures: Procedures A/B pressurize to 14.0 kPa; Procedures C/D use stepped pressurization up to 20.0 kPa-selected to rank material performance.
- Limitations: not fully representative of real-use stresses (e.g., high mechanical pressures, abrasion, flexing, pre-wetting) and may be ineffective for multilayer materials with thick absorbent inner liners.
Applications and who uses ISO 16603
- PPE manufacturers use ISO 16603 to screen and benchmark fabric and seam constructions for blood and body-fluid resistance.
- Independent test laboratories run standardized penetration tests for compliance, quality control, and product certification support.
- Healthcare procurement and infection-control professionals reference test results to select garments that reduce exposure risks to blood-borne pathogens.
- Regulatory bodies and standards committees use the method as part of performance specifications and in comparative assessments.
Related standards
- ISO 13994 - penetration test apparatus specification (used by ISO 16603)
- ISO 16604 - viral penetration resistance (complementary, more sophisticated viral tests)
- ISO 2859-1 - sampling plans for quality control (referenced for larger data sets)
- ASTM F1670 - original basis for ISO 16603
ISO 16603 is intended for comparative material evaluation and quality assurance; users should consider additional preconditioning and complementary tests (e.g., laundering, abrasion, viral testing) when assessing real-world protective performance.
