ISO 16720:2005 specifies a method for pretreatment of soil samples by freeze-drying for subsequent analysis. ISO 16720:2005 is applicable to soil samples for subsequent determination of elements or organic compounds recognized as non-volatile under freeze-drying conditions. Generally, ISO 16720:2005 can also be applied to samples from sludges and sediments. This method is also applicable as a first step for the determination of dry matter (or water) content, for instance in the case of samples with high water content.
Abstract
Overview
ISO 16720:2005 is an international standard that specifies the pretreatment of soil samples by freeze-drying to prepare them for subsequent analytical testing. Developed by the International Organization for Standardization (ISO), this standard provides a reliable and efficient method to remove moisture from soils, sludges, and sediments while preserving non-volatile elements and organic compounds for accurate laboratory analysis.
This method is particularly designed for samples that contain high water content and is also applicable as a first step in determining dry matter or water content. Freeze-drying, also known as lyophilization, helps maintain sample integrity by sublimating water under controlled low temperature and vacuum conditions, preventing alteration of the compounds of interest.
Key Topics
-
Scope and Applicability
The standard applies primarily to soil samples but can also be extended to sludges and sediments. It targets the preservation of elements and organic compounds that remain stable during freeze-drying. -
Freeze-Drying Principle
Freeze-drying involves:- Pre-cooling samples below their eutectic point (usually < −40 °C) to ensure complete freezing.
- Sublimation of ice under vacuum, where ice turns directly into vapor without passing through liquid phase.
- Condensation and collection of moisture on a cooled condenser at temperatures below −50 °C.
- Controlled heat supply to enable sublimation without thawing samples or losing volatile compounds.
-
Sample Preparation
- Samples must be spread in thin layers not exceeding 2 cm to ensure effective drying.
- Containers like trays or flasks are selected based on sample quantity and the freeze-dryer type.
- Hygienic handling is required due to the dust formation from freeze-dried samples.
-
Dry Matter Determination
- Freeze-dried samples are considered partly dried; further drying at 105 °C following ISO 11465 is recommended for precise dry matter or water content analysis.
- The standard outlines formulas to calculate dry matter content accounting for residual moisture and mass differences before and after freeze-drying.
-
Equipment Requirements
- Thermally isolated drying chambers with temperature-controlled carriers or external inlet manifolds.
- Vacuum systems, condensers, and freezers capable of reaching −35 °C or lower.
- Sample containers compatible with freeze-drying apparatus.
-
Avoiding Sample Degradation
- Freeze-drying settings must prevent thawing to avoid loss of non-volatile compounds.
- Certain volatile substances, like ammonia nitrogen in sludges, may be lost, affecting analysis results.
- Sample protection techniques, such as wire meshes, minimize loss of fine particles during vacuum operation.
Applications
ISO 16720:2005 is critical in environmental science, agriculture, and geotechnical engineering where soil quality and composition are analyzed. Key applications include:
-
Soil Quality Assessment
Preparing soil samples to measure elemental content and organic compounds for pollution monitoring, fertility studies, or remediation projects. -
Sediment and Sludge Analysis
Applying freeze-drying for samples with high water content, especially when volatile loss must be minimized. -
Laboratory Sample Preparation
Enabling subsequent physico-chemical or organic compound analyses by ensuring samples are dry, stable, and representative. -
Dry Matter and Water Content Determination
Serving as a preparatory step to accurately measure moisture and dry matter in environmental and industrial samples.

ISO 11465: Soil Quality - Determination of Dry Matter and Water Content on a Mass Basis - Gravimetric Method

By standardizing the freeze-drying pretreatment of soil and related samples, ISO 16720:2005 ensures consistent, reliable, and accurate analytical outcomes fundamental to environmental monitoring and research. Employing this standard enhances laboratory efficiency while protecting the integrity of sensitive sample components.
