ISO 18330|IDF 188:2003 gives guidelines for the standardized description of immunoassays or receptor assays for the detection of antimicrobial residues in milk and milk products. It is intended to provide a framework and basis for the evaluation/validation of tests based on the binding of an antimicrobial compound to its specific antibody or to other types of detecting molecules. In addition to immunoassays [e.g. enzyme-immunoassay (EIA) and radio-immunoassay (RIA)], there are several quantitative, semi-quantitative and qualitative test formats based on the binding of antimicrobial compounds to microbial receptors or to receptor proteins. Enzymatic assays and particle-based assays based on receptor proteins are referred to as receptor assays in ISO 18330|IDF 188:2003.
Abstract
Overview
ISO 18330:2003 (IDF 188:2003) provides guidelines for the standardized description, evaluation and validation of immunoassays and receptor assays used to detect antimicrobial residues in milk and milk products. It covers test formats based on binding of antimicrobial compounds to specific antibodies or receptor molecules - including enzyme-immunoassay (EIA), radio-immunoassay (RIA), enzymatic receptor assays and particle-based receptor protein assays - and establishes a framework for reporting method characteristics and performance.
Key topics and technical requirements
- Scope and method types: Describes quantitative, semi‑quantitative and qualitative formats (EIA, RIA, receptor protein assays).
- Definitions and performance metrics: Standardized meanings for false positives, false negatives, specificity, and limits of detection (qualitative and quantitative).
- Information from developer/manufacturer: Required documentation on method principle, sample preparation, evaluation criteria (visual vs instrumental), capacity, field of application, and known limitations.
- Test kit reagent details: Antibody types (monoclonal/polyclonal), antigen formats, markers (enzymes, radiolabels, latex beads), receptor types and labelling information.
- Operating requirements: User training, laboratory equipment (microplate readers, fluorometers, incubators, pipettes), safety (radioactive handling) and quality assurance procedures.
- Validation and evaluation: Guidance on experimental design (preferably blind coded), number of replicates (typically 10–20 per concentration level), preparation of “negative milk”, selection of test concentrations, dilution/preparation precautions and preservation (lyophilization, deep-freezing).
- Documentation expectations: User manuals, troubleshooting, supplier contacts, availability of reference materials and status of official recognition.
- Legal and practical limits: Notes that many jurisdictions require positive immunoassay/receptor assay results to be confirmed by an accepted physico‑chemical method.
Applications and users
ISO 18330 is intended for:
- Test developers and manufacturers documenting and validating immunoassays and receptor assays.
- Dairy testing laboratories performing routine quality control, screening for antimicrobial residues, and post‑screening identification.
- Regulatory agencies and food safety authorities designing surveillance programs and interpreting screening data.
- Research and proficiency testing organizations standardizing methodology and inter-laboratory studies.
Practical applications include primary screening for milk quality or regulatory compliance, rapid on-site testing, and selection/validation of assays for detecting residues at or around maximum residue limits (MRLs).
