ISO 20481:2008 specifies a high performance liquid chromatography (HPLC) method for the determination of the caffeine content of: green coffee; roasted coffee; soluble coffee, regular and decaffeinated; and mixed instant coffee products (e. g. coffee/chicory mix or cappuccino-type coffee drink).
Abstract
Overview
ISO 20481:2008 - "Coffee and coffee products - Determination of the caffeine content using high performance liquid chromatography (HPLC) - Reference method" defines a validated HPLC reference procedure for measuring caffeine in green coffee, roasted coffee, soluble (instant) coffee - regular and decaffeinated - and mixed instant coffee products (e.g., coffee/chicory blends, cappuccino-type powders). The method specifies sample preparation, extraction, chromatographic conditions and calculation rules to deliver traceable, repeatable caffeine results.
Key topics and technical requirements
- Sample scope: Green beans, roasted beans, soluble coffee, decaffeinated and mixed instant products.
- Extraction: Hot-water extraction at ≥90 °C with magnesium oxide (MgO) to protect column life; typically 5 g MgO and about 200 ml water per test portion, heated 20 minutes.
- Chromatography: Reversed-phase C18 column (≥125 mm, ~5 µm, ≥5 000 theoretical plates), isocratic elution with a mobile phase of 24% (v/v) methanol in water.
- Detection and injection: UV detection at ~272 nm (270–280 nm acceptable), typical flow rate 1.0 ml/min and injection volume 10 µl.
- Standards and calibration: Stock caffeine standard 200 mg/l with working dilutions (≈40 mg/l for regular, ≈4 mg/l for decaf); 3–5 point calibration graph optional.
- Quality and safety: Use HPLC-grade reagents and ISO 3696 grade 1 water; degas mobile phase; safety measures for methanol handling (gloves, eye protection, fume hood).
- Results and reporting: Formulae for as‑is and dry‑basis caffeine (% mass) provided; report must reference ISO 20481 and include test details and any deviations.
- Precision: Repeatability and reproducibility data from interlaboratory studies are supplied (Annex A / Table 1) to guide interpretation of r and R limits.
Applications and who uses it
- Industrial QC laboratories at coffee roasters, instant coffee manufacturers and ingredient suppliers for product specification, batch release and supplier control.
- Testing and certification bodies using a recognized reference method for regulatory compliance, labeling claims (caffeine content) and dispute resolution.
- Research and R&D teams studying caffeine extraction, decaffeination processes or product formulation (mixes, beverages).
- Customs/import labs and food safety authorities requiring harmonized methods for enforcement and trade.
