This document specifies a method for the quantification of selected volatile organic compounds (VOCs: 1,3-butadiene, isoprene, acrylonitrile, benzene and toluene) by GC-MS in mainstream cigarette smoke using ISO 3308 smoking parameters. This method is applicable to cigarettes with nicotine-free dry particulate matter (NFDPM) yields between 1 mg/cigarette and 15 mg/cigarette.
Abstract
Overview
ISO 21330:2018 specifies a validated laboratory method for quantifying selected volatile organic compounds (VOCs) in the mainstream smoke of cigarettes using GC–MS. The standard targets five VOCs - 1,3‑butadiene, isoprene, acrylonitrile, benzene and toluene - collected under the ISO 3308 smoking regime and applies to cigarettes with NFDPM (nicotine‑free dry particulate matter) yields between 1 mg and 15 mg per cigarette. The method uses cryogenically cooled impinger traps with methanol, internal standards (e.g., benzene‑D), and GC–MS analysis performed in Selected Ion Monitoring (SIM) mode.
Key topics and technical requirements
- Scope & analytes: 1,3‑butadiene, isoprene, acrylonitrile, benzene, toluene in mainstream cigarette smoke.
- Smoking parameters: Machine smoking must comply with ISO 3308 (standard puff profile and conditions).
- Sampling: Mainstream smoke is passed through a Cambridge filter pad into series cryogenic impingers containing methanol for vapour‑phase trapping.
- Trapping & solvents: Use of cryogenic (dry‑ice cooled) impingers and HPLC‑grade methanol; methanol must be checked for background analyte levels.
- Standards & calibration: Preparation of primary and secondary stock solutions, internal standard spiking (benzene‑D recommended), calibration and calculation procedures are defined. Storage guidance (e.g., frozen vials) is included.
- GC–MS setup: Split injection on a capillary column (example: DB‑624, 60 m × 0.25 mm × 1.4 μm), SIM acquisition recommended; auto‑sampler suggested.
- Quality & performance: Repeatability and reproducibility data from collaborative studies (including a 2012 study) and statistical evaluation per ISO 5725 series.
- Safety & limitations: The method involves hazardous materials and equipment; machine smoke yields are not direct measures of human exposure or risk.
Applications and users
- Who uses it: Analytical laboratories, tobacco product testing labs, regulatory agencies, research institutes, and quality control groups in tobacco industry and public health research.
- Practical applications:
- Routine monitoring and reporting of VOC yields in cigarette mainstream smoke.
- Product development and formulation comparison.
- Regulatory compliance testing and inter‑laboratory comparability.
- Input data for hazard assessment (with caveats about not equating machine yields to human exposure).
