Abstract
Overview
ISO 3400:1997 specifies a spectrometric method for the determination of alkaloids, primarily nicotine, in smoke condensates from cigarettes. Developed by ISO Technical Committee 126 (Tobacco and tobacco products), this international standard provides a detailed, reproducible approach to quantify total alkaloid content, supporting quality control and regulatory compliance within the tobacco industry. This third edition replaces previous iterations, refining procedures for increased accuracy and reliability.
Key Topics
- Alkaloid Measurement in Smoke: The method outlined quantifies total alkaloids, expressed as nicotine, in cigarette smoke condensates obtained through standardized or non-standard smoking protocols.
- Spectrometric Procedure: Utilizes dissolution, steam distillation, and spectrometric absorbance measurement at specific wavelengths (236 nm, 259 nm, 282 nm) to achieve precise results.
- Laboratory Requirements: Specifies reagent purity (analytical grade solvents and water per ISO 3696), apparatus (steam distillation system, spectrometer, matched quartz cells), and the need to validate instrument performance regularly.
- Sample Preparation: Details steps for preparing smoke condensates, ensuring accurate representation of cigarette yields.
- Reporting: Outlines requirements for test reports to include nicotine yield per cigarette and all relevant test conditions for traceability.
Applications
- Tobacco Industry Quality Assurance: Standardized measurement of nicotine and other alkaloids ensures consistency and compliance with local and international regulations.
- Product Development: Enables manufacturers to monitor smoker exposure to nicotine, supporting product innovation and harm reduction efforts.
- Regulatory Compliance: Offers a recognized method for official control laboratories and regulators when assessing nicotine yields, especially in jurisdictions without access to gas-chromatography technology.
- Comparative Analysis: Facilitates comparison between products or smoking regimes by providing repeatable and internationally accepted measurement data.
- Research and Development: Provides a foundation for scientific investigation into the chemical analysis of cigarette smoke, supporting broader tobacco control studies and public health initiatives.
Related Standards
- ISO 4387: Specifies methods for determining total and nicotine-free dry particulate matter using smoking machines, referenced for standard smoking protocols.
- ISO 10315: Details the gas-chromatographic method for nicotine determination, offering an alternative technique focused solely on nicotine rather than total alkaloids.
- ISO 13276: Establishes procedures for determining nicotine purity using a gravimetric method with tungstosilicic acid, supporting reagent verification.
- ISO 3696: Specifies requirements for water purity in analytical laboratory applications, ensuring consistent testing conditions.
- ISO/IEC Guide 25: Sets general requirements for the competence of calibration and testing laboratories, underpinning methodological reliability.
By following ISO 3400:1997, laboratories and industry stakeholders can ensure accurate, reproducible measurement of alkaloid content in cigarette smoke, supporting regulatory submissions, product quality management, and scientific research. Integrating this standard into laboratory protocols enhances trust in analytical results and aligns practices with global tobacco testing norms.
