Abstract
Overview
ISO 3596:2000 specifies a laboratory method for the determination of unsaponifiable matter in animal and vegetable fats and oils using diethyl ether extraction. The procedure involves saponifying a weighed test portion with ethanolic potassium hydroxide, extracting the unsaponifiable fraction with freshly distilled diethyl ether, evaporating the solvent, drying the residue and reporting the result as a percentage by mass of the sample. The method is not applicable to waxes and can give approximate results for some high‑unsaponifiable fats (for example, certain marine oils).
Key topics and technical requirements
- Scope & limitations: Applicable to most animal and vegetable fats and oils; not suitable for waxes; alternative method (hexane extraction) available in ISO 18609 when diethyl ether cannot be used.
- Principle: Saponification with ethanolic KOH followed by extraction of non‑saponifiable components in diethyl ether, evaporation and gravimetric determination.
- Reagents & apparatus:
- Diethyl ether (freshly distilled, free from peroxides), acetone, ethanolic and aqueous KOH, phenolphthalein.
- Standard glassware: 250 ml round‑bottom flasks, reflux condenser, 500 ml separating funnel, boiling water bath, oven at 103 °C.
- Procedure highlights:
- Test portion ≈ 5 g; reflux saponification for 1 hour.
- Triple extraction with 100 ml diethyl ether; successive washings until neutral (phenolphthalein).
- Evaporate ether, add acetone to remove residues, dry residue at 103 °C to constant mass.
- Optional titration to correct for free fatty acids.
- Calculation & reporting: Result expressed as % (mass) of unsaponifiable matter after blank correction and optional free fatty acid correction; two determinations recommended.
- Precision: Annex A summarizes interlaboratory studies (soybean and fish oils) giving repeatability and reproducibility statistics - useful for interpreting method variability.
Applications and users
ISO 3596:2000 is used by:
- Food and oil analysis laboratories for routine quality control of edible oils and fats.
- Oil processors and refiners monitoring refining performance (removal of unsaponifiables).
- Regulatory and conformity testing bodies assessing product composition.
- Research labs analyzing tocopherols and other unsaponifiable constituents (note: for tocopherol composition, pyrogallol addition and rapid extraction are recommended). Users should be trained in solvent handling and laboratory safety (diethyl ether is flammable and prone to peroxide formation).
