As measurements of the degree of wall thickening of the fibres is too laborious for most practical purposes, this method of determination of maturity of cotton fibres is an indirect test. It consists of an appraisal, based on judgment and experience, and is suitable for routine research purposes.

ISO 4912:1981 - Textiles - Cotton fibres - Evaluation of maturity - Microscopic method defines an indirect, visual procedure to evaluate the maturity of cotton fibres by microscopic examination after chemical swelling. The method is an appraisal based on trained judgment and experience rather than direct wall-thickness measurements, and is intended for routine research and quality-control applications in textile testing.

- Option 1: classify fibres into dead, normal, thin‑walled and compute a maturity ratio from mean percentages of normal and dead fibres (formula provided in the standard).
- Option 2: classify fibres as immature or mature and calculate percent maturity = (mature fibres / total fibres) × 100.
- Annex C contains a conversion table and equations to interconvert maturity ratio and percent maturity.

- Test report must reference ISO 4912, state specimen preparation method, give measured maturity (ratio or percent), and note any deviations or incidents.
