Abstract
Overview
ISO 5565-2:1999, "Vanilla [Vanilla fragrans (Salisbury) Ames] - Part 2: Test methods", specifies analytical methods for the determination of key quality parameters in vanilla pods and powder. It is applicable to vanilla in pods (cut or bulk) and powdered form but is not applicable to vanilla extracts. This Part 2 complements ISO 5565-1 (Specification) and was prepared by ISO/TC 34 (Agricultural food products), SC 7 (Spices and condiments).
Key topics and technical requirements
- Scope and sample preparation
- Applies to Vanilla fragrans (syn. Vanilla planifolia) in pods and powder; pods should be cut into pieces (max 5 mm) and powders homogenized prior to testing.
- Moisture determination (azeotropic distillation)
- Uses toluene saturated with water; azeotropic distillation and graduated receiver (5 ml capacity, 0.10 ml divisions).
- Typical test portion ~10 g; apparatus includes a 500 ml flask, reflux condenser, 5 ml graduated receiver and heat source (oil bath/electric heater).
- Moisture content w (%) = 100 × V / m, assuming water density 1 g/ml.
- HPLC reference method for vanillin and related compounds
- Targets: vanillin, vanillic acid, 4-hydroxybenzaldehyde, 4-hydroxybenzoic acid.
- Sample extraction: Soxhlet/continuous extraction with ethanol (≈200 ml) for ~16 h; final extract diluted to 200 ml and typically 1:10 diluted in mobile phase.
- Chromatography: mobile phase guidance is dilute phosphoric acid : methanol (75:25), C18 bonded silica column (5–10 μm), UV detection at 254 nm.
- Quantification uses an internal standard (acetylsalicylic acid), response coefficients (peak height or area), and calibrated stock/working solutions.
- Filtration through 0.45 μm membranes and injection volumes 10–100 μl.
- UV spectrometric method for vanillin
- Vanillin extracted with ethanol and assayed spectrometrically (UV), using appropriate reagents (e.g., NaOH solution referenced) and standard preparation.
Applications and users
- Quality control laboratories in the spice, food flavoring and food ingredient industries performing routine and reference analyses.
- Vanilla producers, packers and trading houses verifying moisture and vanillin content for specification compliance.
- Regulatory and standards organizations, research laboratories and contract testing services requiring standardized, reproducible test methods for vanilla raw materials.
- Useful for product specification, lot release testing, and comparative analytics between HPLC (reference) and faster UV screening.
