ISO 9509:2006 specifies a method for assessing the short-term inhibitory effect of waters, waste waters or test substances on nitrifying bacteria in activated sludge. The inhibitory effect is estimated over an exposure period of usually 3 h or up to 24 h with weakly nitrifying sludge. The method is applicable to nitrifying activated sludge derived from domestic and synthetic sewage and also to sludges from industrial and mixed domestic and industrial waste waters. The nitrifying activity of the sludge is verified by testing in the presence and absence of a specific inhibitor. If the nitrification rate is within a suitable range for the test, i.e. 2 mg of nitrogen per gram of suspended solid and hour to 6,5 mg of nitrogen per gram of suspended solids and hour, the sludge may be used directly. If not, adjustments are necessary. The method is applicable to water-soluble, non-volatile chemicals, and to waste waters
Abstract
Overview
ISO 9509:2006 - Water quality: Toxicity test for assessing the inhibition of nitrification of activated sludge microorganisms specifies a laboratory method to determine short‑term inhibitory effects of waters, wastewaters or test substances on nitrifying bacteria in activated sludge. The test estimates inhibition by measuring oxidized nitrogen (nitrite + nitrate) production or ammonium removal over a typical exposure of 3 hours (up to 24 hours for weakly nitrifying sludge). The method applies to nitrifying activated sludge from domestic, industrial or mixed sewage and to water‑soluble, non‑volatile chemicals and wastewaters.
Key topics and technical requirements
- Measured endpoints: production of oxidized nitrogen (NO2– + NO3–) and/or disappearance of ammonium‑N (NH4‑N).
- Exposure time: usually 3 h (can be extended to 24 h for weakly nitrifying sludges).
- Sludge activity verification: test sludge must have nitrification rates in the recommended range 2 to 6.5 mg N · (g SS)⁻¹ · h⁻¹; Annex A describes activity determination and adjustments.
- Reference inhibitor: N‑allylthiourea (ATU) is used to verify nitrifying activity and as a control.
- Test conditions: typical incubation at 22 °C ± 2 °C, aeration or shaking to maintain dissolved oxygen > 4 mg/L, dark or diffused light. Reaction vessels include conical flasks or measuring cylinders.
- Media & reagents: defined bicarbonate/ammonium media (composition provided in the standard) to supply ammonium‑N (~56 mg/L when diluted 1:10).
- Result expression: percent inhibition and EC50 (effective concentration causing 50% inhibition) are calculated by comparing treated and control nitrification rates.
- Safety note: activated sludge and sewage can contain pathogens; tests must be run by trained staff following laboratory safety procedures.
Applications and who uses it
- Environmental laboratories and ecotoxicologists for screening chemical toxicity to nitrifying bacteria.
- Wastewater treatment plant (WWTP) operators and process engineers for diagnosing nitrification inhibition events and troubleshooting treatment performance.
- Regulatory agencies and consultants performing wastewater discharge impact assessments and compliance testing.
- Researchers developing or validating toxicity mitigation measures and biodegradability studies involving nitrifying microorganisms.
