La présente Norme internationale spécifie une méthode pour la détermination du pourcentage, en volume, de phénols dans les huiles essentielles. NOTE :Voir en 8.2.2, où sont établies certaines limites d'application de la méthode.
Abstract
Overview
ISO 1272:2000 - Essential oils - Determination of content of phenols - specifies a standardized volumetric procedure for measuring the phenol content (percentage by volume) in essential oils. The method converts phenolic compounds to their water‑soluble alkaline phenates and determines the volume of oil remaining unabsorbed. ISO 1272:2000 is the second edition, replacing the 1973 version, and is intended for laboratory quality control and analytical verification.
Key topics and technical requirements
- Principle: Phenols in a known volume of essential oil are transformed into alkaline phenates (soluble in aqueous phase); unabsorbed oil volume is read and converted to phenol content.
- Reagents: tartaric acid (pretreatment), potassium hydroxide (1 mol/L, silica‑ and alumina‑free), and xylene (for resolving emulsions). Use analytical‑grade reagents and purified water.
- Apparatus: one‑mark pipettes (2 ml, 10 ml, ISO 648), 100 ml conical flasks (ISO 1773), and a 150 ml Cassia flask with a 10 ml graduated neck (0.1 ml divisions).
- Sample preparation: follow ISO 356 for test sample preparation; pretreat with tartaric acid to avoid interphase emulsions and dry (magnesium sulfate per ISO 356).
- Procedure highlights:
- Use 10 ml prepared sample with ~75 ml KOH solution, shake six times at 5‑minute intervals (standard temperature 20 °C unless specified otherwise).
- Add extra KOH until unabsorbed oil rises into the Cassia flask neck; read volume after standing.
- If emulsions persist, add 2 ml xylene (or pre‑add before shaking) to break emulsions.
- Some oils (containing phenol esters) may require heating or modified shaking-monograph‑specific instructions apply.
- Calculation: phenol content w (%) = 10^(10 − V), where V is the unabsorbed oil volume in ml (subtract 2 ml if xylene was added). Report to the nearest whole number.
- Performance: repeatability - in not more than 5% of cases, two independent results will differ by more than 1% (absolute).
Applications and users
ISO 1272:2000 is used by:
- Quality control and analytical laboratories in the essential oils, flavour and fragrance, cosmetics, and natural products industries.
- Regulatory and pharmacopoeial testing bodies verifying phenol content for safety, labeling, and specifications.
- Producers and exporters assessing batch conformity and stability.
Practical benefits include reproducible phenol quantification for product specification, safety assessment (phenolic content relates to irritation/allergen potential), and comparative quality control across suppliers.
