ISO 21415-2:2015 specifies a method for determining the content of wet gluten and the gluten index for wheat flours (Triticum aestivum L. and Triticum durum Desf.) by mechanical means. This method is directly applicable to flours. It also applies to common and durum wheat after grinding, if their particular size distribution meets the specification given in Table B.1.

ISO 21415-2:2015 specifies a laboratory mechanical method for determining wet gluten content and the gluten index in wheat flours (Triticum aestivum L. and Triticum durum Desf.). The standard applies directly to flours and to ground whole wheat when the particle-size distribution meets the normative requirement (Table B.1). The method isolates wet gluten by forming a paste with a sodium chloride solution, washing out starch and soluble components, then centrifuging and weighing the gluten. The gluten index is defined as the percentage of wet gluten remaining on a sieve after centrifugation.
