Liquid chromatography is a method of separation based on the phenomena of adsorption, partition, ion exchange and/or exclusion. It enables a small quantity of aromatic essential oil or other raw materials for the perfume industry to be analysed on a chromatographic column with an appropriate packing and under appropriate conditions, the possible identification of the different constituent and the quantitative determination of specific compounds by measuring the area or height of their peaks. Details of the method are described.
Abstract
ISO 8432:1987 - Essential Oils Analysis by High Performance Liquid Chromatography
Overview
ISO 8432:1987 specifies a general method for analyzing essential oils using High Performance Liquid Chromatography (HPLC). This standard outlines the procedure for separating, identifying, and quantifying specific compounds within aromatic essential oils and related raw materials for the perfume industry. The method leverages key chromatographic principles such as adsorption, partition, ion exchange, and exclusion to achieve precise component resolution.
The primary objective of ISO 8432:1987 is to enable the reliable determination of the chemical composition of essential oils via an articulated chromatographic approach, providing essential data on individual constituents by measuring peak area or height in the chromatogram.
Key Topics
- Principle of HPLC for Essential Oils: Uses specialized chromatographic columns with suitable packing materials and detectors to separate and identify oil constituents.
- Reagents and Materials: Includes purified reference substances for calibration, internal or external standards, solvents, and inert gases like helium or nitrogen for system operation.
- Apparatus Requirements:
- Chromatograph with pumping and solvent supply systems, capable of delivering constant or programmed solvent flow.
- Inert columns made of glass or stainless steel that withstand operational pressures.
- Detection systems compatible with quantification demands.
- Sample Preparation: Involves techniques outlined in ISO 356 or other relevant ISO standards to prepare injection-ready test samples ensuring particle-free solutions.
- Chromatographic Procedure:
- Control of elution solvent flow rate and temperature to optimize constituent separation.
- Ability to operate in isocratic (constant mobile phase composition) or gradient elution modes.
- Quantitative Determination Methods:
- Internal Standard Method – uses a substance with a retention time close to the target compound to improve precision.
- Addition and External Standard Methods – alternative approaches for cases where internal standards are unsuitable.
- Performance Metrics and Calculations:
- Assessment of column efficiency via theoretical plate number.
- Calculation of resolution between neighboring peaks for adequate separation.
- Calculation of compound concentrations from chromatogram peak areas using response factors.
- Test Reporting: Comprehensive details required including apparatus specifications, column characteristics, detector settings, mobile phase data, sample identity, and results.
Applications
- Quality Control in Perfume Industry: Ensures essential oils meet compositional specifications and regulatory requirements before their use in fragrance formulations.
- Research and Development: Facilitates analysis of complex oil matrices during new product development or process optimization.
- Authentication and Adulteration Detection: Helps detect impurities or adulterants in essential oils by defining component profiles.
- Regulatory Compliance: Supports conformity checks according to international trade standards for essential oils.
