Abstract
Overview
ISO/TR 13014:2012 - Nanotechnologies: Guidance on physico‑chemical characterization of engineered nanoscale materials for toxicologic assessment - is an ISO Technical Report published in 2012 that provides informative guidance (not normative requirements) for the physico‑chemical characterization of manufactured nano‑objects and their aggregates/agglomerates (NOAA) presented for toxicological testing. The report is intended to help toxicologists and related professionals identify and measure critical properties of test materials so toxicological results can be interpreted, compared and reproduced.
Key Topics
The report covers terminology, measurement concepts and the physico‑chemical parameters important to toxicologic assessment, including:
- Terms and definitions relevant to nano‑objects, nanoparticles, nanofibres, aggregates and agglomerates.
- Importance of physico‑chemical properties for toxicology and when characterization should be undertaken.
- Core characterization parameters (clause 5):
- Particle size and size distribution
- Aggregation/agglomeration state in relevant media
- Shape
- Surface area (mass‑ and volume‑specific)
- Composition and crystallinity
- Surface chemistry and surface charge
- Solubility / dispersibility
- Measurement expression and uncertainty: guidance on measurands, metrological traceability, combined/expanded measurement uncertainty, and validation.
- Reporting requirements and informative annexes: a diagram showing how characterization fits into testing and an annex with example measurement methods and standards.
Practical Applications
ISO/TR 13014:2012 is practical guidance for:
- Toxicologists and ecotoxicologists planning biological tests on manufactured nano‑objects.
- Regulators and risk assessors needing clear, documented material characterization to interpret study outcomes.
- Health & safety professionals and laboratory scientists designing sampling, dispersion and analytical protocols.
- Manufacturers and R&D teams who must differentiate seemingly similar nano‑materials and provide characterization data for safety dossiers.
Use cases:
- Establishing a characterization checklist prior to in vitro or in vivo toxicology studies.
- Documenting particle state (size, aggregation, surface chemistry) to improve reproducibility and enable cross‑study comparisons.
- Supporting regulatory submissions and material safety evaluations.
