This International Standard specifies a method, by analysis of inorganic carbon, for the evaluation in an aqueous medium of the ultimate aerobic biodegradability of organic substances at a given concentration of microorganisms. This International Standard is applicable to organic compounds which are: a) water-soluble under the test conditions; b) poorly water-soluble under the test conditions, in which case special measures may be necessary to achieve a good dispersion of the compound (see ISO 10634); c) volatile; d) not inhibitory to the test microorganisms at the concentration chosen for the test. In this test, biogenically produced inorganic carbon is measured in situ in the test vessels in such a manner that the rate measured nearly equals the rate of microbial production. NOTE 1 The conditions described in this International Standard do not always correspond to the optimal conditions for allowing the maximum degree of biodegradation to occur. For alternative biodegradation methods see ISO 15462. NOTE 2 With highly volatile substances, losses to the gaseous phase can be minimized by reducing the volume of the headspace. However, there should be sufficient oxygen in the test system to prevent biodegradation being oxygen-limited. NOTE 3 The presence of an inhibitory effect can be determined as specified in 8.3, or by using any other method for determining the inhibitory effect of a substance on bacteria (see, for example, ISO 8192).
Abstract
Overview
EN ISO 14593:2005 specifies a standard method for evaluating the ultimate aerobic biodegradability of organic compounds in water by analyzing inorganic carbon in sealed vessels-a process commonly known as the CO₂ headspace test. Developed by the European Committee for Standardization (CEN), this international standard is critical for water quality assessment and the environmental evaluation of chemicals.
This method measures the biochemical conversion of organic substances into inorganic carbon (e.g., CO₂) under controlled aerobic conditions, simulating natural microbial degradation in aquatic environments. It is suitable for a range of organic compounds, including those that are water-soluble, poorly soluble (with special preparation), volatile, or non-inhibitory to the test microorganisms.
Key Topics
- Evaluation of Biodegradability: The standard describes procedures for quantitatively measuring the ultimate biodegradation (mineralization to CO₂) of organic substances in an aqueous medium.
- Test Applicability: Applicable to organic compounds that are fully or partially water-soluble, as well as volatile substances, provided they do not inhibit microorganism activity at tested concentrations.
- CO₂ Headspace Method: The core of the method is the measurement of biogenic inorganic carbon produced in situ within sealed test vessels, ensuring results closely reflect microbial activity.
- Test Conditions: Includes details on medium composition, inoculum origin and preparation (e.g., activated sludge, wastewater, surface water), temperature control, oxygen supplementation, and sample handling for reliable outcomes.
- Result Calculation and Validation: Outlines calculation of biodegradation percentage based on measured inorganic carbon compared to theoretical maximum, as well as criteria for test validity and statistical treatment.
Applications
- Water Quality Monitoring: Used by environmental laboratories and water treatment facilities to assess the environmental impact of chemicals released into surface waters, groundwater, or wastewater systems.
- Regulatory Compliance: Supports industry and regulators in demonstrating that substances meet environmental safety requirements for biodegradability.
- Risk Assessment for New Chemicals: Enables manufacturers and researchers to evaluate the persistence of new organic compounds, aiding in risk assessments and product stewardship.
- Product Development and Labeling: Informs eco-labeling and marketing claims regarding the biodegradability of detergents, surfactants, pharmaceuticals, and other organic products.
- Research and Benchmarking: Essential for scientists studying microbial ecology, environmental chemistry, and the fate of organic pollutants in aquatic systems.
Related Standards
To ensure a comprehensive approach to water quality and biodegradability testing, the following related international standards should be considered:
- ISO 10634: Guidance on preparation and treatment of poorly water-soluble organic compounds for biodegradability testing.
- ISO 15462: Provides alternative methods for evaluating aerobic biodegradability.
- ISO 8192: Determines inhibitory effects of substances on bacterial activity, which can be relevant for selecting appropriate test concentrations.
- ISO 7827 & ISO 8245: Additional methods for assessing the biodegradability of organic compounds in aqueous media.
These standards collectively help laboratories, industry, and regulators systematically evaluate the environmental behavior of a wide range of organic chemicals, safeguarding water quality and public health.
By implementing EN ISO 14593:2005, organizations ensure consistent, reliable assessments of organic compound biodegradability, supporting environmental protection, regulatory compliance, and informed decision-making in water quality management.
