ISO 16712:2005 specifies a method for the determination of acute toxicity to amphipods exposed over a period of 10 d to samples of contaminated marine or estuarine sediment, to chemical, industrial or municipal sludge or other solid wastes that may combine with marine or estuarine sediments, or to chemicals or preparations spiked into clean sediment.

EN ISO 16712:2006 (ISO 16712:2005) is a CEN-adopted international standard for water quality testing that specifies a laboratory method to determine the acute toxicity of marine or estuarine sediments to amphipods. The method covers exposure of sediment-dwelling amphipods to contaminated sediments, sludges or sediments spiked with chemicals, with a biological endpoint based on percent mortality after a 10‑day exposure. The standard is intended for environmental monitoring, risk assessment and sediment quality evaluation.

Test vessels & media: Tests are performed in controlled laboratory vessels (standard 1‑litre type is specified in the standard) containing a solid-phase sediment layer and overlying water. Both field-collected sediments and sediments spiked with test chemicals are allowed.

Test environment: Facilities must be free from contamination and disturbance. Lighting is uniform and continuous to prevent nocturnal amphipod migration; temperature and salinity are controlled to suit the species (see Annex B).

Sample handling: Procedures cover collection, sieving, transport, holding/acclimation, and preparation of control or reference sediments. Species-specific maximum sizes, optimal salinity and temperature ranges are provided in Annex B.

Quality control & validity: The standard includes guidance on controls, reference toxicants, reburrowing capability, observation schedules, data expression and test validity criteria.

Annexes: Normative and informative annexes address reconstituted salt water (Annex A), species parameters (Annex B) and precision data (Annex C).
