Overview

ISO 12966-1:2014 provides guidelines for the gas chromatographic (GC) determination of fatty acids in animal and vegetable fats and oils after they have been converted to fatty acid methyl esters (FAMEs). Part 1 is a high-level guideline to modern gas chromatography of FAMEs, describing principles, column selection, detection choices and chromatogram evaluation. It complements Parts 2–4, which cover FAME preparation methods and detailed analytical conditions.

Covers qualitative and quantitative GLC/GC analysis of free and bound fatty acids (FAMEs), typically C4–C26, including saturated, cis/trans mono- and polyunsaturated FAMEs.
Not applicable to polymerized or oxidized fats.

Preparation of FAMEs

FAME preparation is performed according to ISO 12966-2 and ISO 12966-3 (different esterification/transmethylation methods such as KOH, BF3, methanolic H2SO4/HCl and TMSH are discussed in the suite).
Selection of esterification method affects conversion of free fatty acids, formation of artefacts and suitability for short-chain fatty acids.

Chromatographic system and detection

Capillary GC with Flame Ionisation Detector (FID) is the standard detection; hydrogen is recommended as carrier gas for FID, helium for mass spectrometric detection (MSD).
Detector choice and carrier gas influence sensitivity, peak shape and analysis time.

Column selection

Wall-coated open tubular (WCOT) capillaries are state-of-the-art. Column lengths typically range 10–100 m, internal diameters from 0.10–0.53 mm, and film thickness 0.1–0.3 μm.
Highly polar cyanoalkyl polysiloxane phases (e.g., SP-2560, CP-Sil88) are recommended for separating geometrical/positional isomers and cis/trans isomers; other phases (BPX70, DB-23, Rtx-2330, etc.) may be used but can change elution order.
Longer columns increase resolution (but with diminishing returns) and may be required for full cis/trans separation.

Chromatogram evaluation

Procedures for peak area, area percent, and use of internal standards or correction factors are included for quantitative work.
