ISO 16373-3:2014 specifies a method for the detection and quantitative determination of the presence of carcinogenic dyestuffs in dyed, printed or coated textile products by chromatographic analysis of their extracts.

Scope: Applies to dyed, printed or coated textile articles for the listed carcinogenic dyestuffs (examples include Basic Red 9, Disperse Orange 11, Disperse Yellow 3, Acid Red 114 - see Table 1 of the standard for the full list).
Extraction method: Use 0.25% triethylamine in methanol; ultrasonic extraction at 50 °C (±2 °C) for 3 hours on a specimen ≤1.0 g. Optional hexane cleansing for grease/oil removal.
Sample concentration & preparation: Evaporate extract (rotary evaporator at ~40 °C), reconstitute residue in 1 ml methanol, filter (0.45 μm PTFE) and dilute if necessary.
Chromatographic analysis: HPLC with diode-array detector (HPLC-DAD) or mass spectrometer (HPLC-MSD). Annex A provides validated HPLC/DAD parameters (e.g., Inertsil ODS-3 column, 0.8 ml/min flow, ammonium acetate/acetonitrile eluents, injection 5 μl) and recommended detection wavelengths for specific dyes.
Calibration & quantification: Use at least four calibration points (recommended 1–100 μg/ml range); linear correlation coefficient ≥ 0.99. Results expressed as mass fraction (μg/g).
Safety: Target dyestuffs are known or suspected carcinogens - follow national health & safety regulations and good laboratory practice.
Test report elements: Reference to ISO 16373-3, specimen details, methods used, results (levels and detection limits in μg/g), and any procedural deviations.
