ISO 19056-1:2015 specifies procedures for the measurement of image brightness and uniformity for bright field light microscopy. These measurements for image brightness and uniformity are defined in image planes or intermediate image planes only, when these planes are suitable for detection by electronic imaging devices. ISO 19056-1:2015 defines how image brightness and uniformity are measured and how this information is provided to the user. NOTE The scope is intentionally limited to electronic imaging devices and (intermediate) image planes. The visual observation by means of eyepieces would require a different measurement procedure and hence result in unambiguities in the description of measurement procedures. Nevertheless, this part of ISO 19056 will give useful estimates for the image brightness with visual observation as in this case, an eyepiece is used to observe an intermediate image plane (which is under the scope of this part of ISO 19056).
Abstract
Overview
ISO 19056-1:2015, titled Microscopes - Definition and measurement of illumination properties - Part 1: Image brightness and uniformity in bright field microscopy, is an international standard established by ISO to define precise procedures for measuring image brightness and uniformity in bright field light microscopy. This part of ISO 19056 focuses exclusively on measurements made at image or intermediate image planes suitable for electronic imaging devices, ensuring consistent and unambiguous evaluation of illumination properties. The standard does not cover direct visual observations through eyepieces but provides useful approximations relevant to these observations.
Accurate measurement of brightness and uniformity is critical for achieving optimal image quality in light microscopy, impacting a wide range of scientific and industrial applications. The standard offers a robust framework for assessing and reporting illumination characteristics to users, supporting improved microscope performance and comparability.
Key Topics
-
Measurement Scope ISO 19056-1 applies to bright field microscopy and covers transmitted or reflected light and various illumination sources, including conventional lamps and lasers. It specifically targets the measurement of brightness (illuminance and irradiance) and uniformity on image planes detectable by electronic sensors.
-
Image Brightness Brightness is quantified using SI units: illuminance (lm/m²) for continuous visible spectral range light sources like halogen lamps and phosphor-type LEDs, and irradiance (W/m²) for other sources such as lasers or narrow-band LEDs. Ensuring sufficient image brightness helps reveal fine object details crucial for microscopy.
-
Uniformity of Illumination Uniformity measures spatial consistency of brightness across the image field, expressed as a percentage ratio of minimum to maximum brightness. Adequate uniformity prevents misinterpretation of specimen features and preserves image fidelity.
-
Measurement Procedures The standard specifies how to set microscope diaphragms to minimize measurement ambiguities, e.g., opening field diaphragms to encompass the image field and matching illumination aperture diaphragms with the objective’s numerical aperture. Brightness is measured via an integrating sphere positioned centrally on the image plane, while uniformity requires an electronic image sensor capturing at least 50x50 pixels to assess brightness distribution. Gamma correction for uniformity measurements must be disabled to maintain signal linearity.
-
User Information and Reporting Results provided to users must include comprehensive information such as image field dimensions and shape, microscope configuration details, illumination source parameters, aperture settings, and spectral ranges applicable to the measurements. This thorough reporting supports reproducibility and user understanding.
Applications
-
Microscope Manufacturers Use ISO 19056-1 to standardize illumination performance testing of bright field microscopes, enabling consistent product evaluation and quality control.
-
Laboratories and Research Facilities Apply the standard for calibrating microscopes to ensure reliable brightness and uniformity during critical imaging tasks, improving accuracy in scientific analysis.
-
Industrial Inspection and Quality Assurance Enhance automated microscopy systems with precise illumination measurements to detect defects or features with higher confidence.
-
Educational and Clinical Settings Ensure microscope illumination meets established criteria, optimizing visual clarity for teaching and diagnostic applications.
