Overview
EN ISO 16140-4:2020/A1:2024 is an amendment to ISO 16140-4:2020 published by CEN/ISO. It provides a single‑laboratory protocol for validating a larger test portion size for qualitative microbiological methods in the food chain. The amendment adds a normative Annex H and clarifies procedures for demonstrating that increasing the analysed test portion does not worsen the method’s level of detection (LOD).

- Pooling rules follow ISO 6887‑1; pooling (test portion) vs pooling (pre‑enrichment) cannot be combined in a single validation-separate studies are needed.
