ISO 9509:2006 specifies a method for assessing the short-term inhibitory effect of waters, waste waters or test substances on nitrifying bacteria in activated sludge. The inhibitory effect is estimated over an exposure period of usually 3 h or up to 24 h with weakly nitrifying sludge. The method is applicable to nitrifying activated sludge derived from domestic and synthetic sewage and also to sludges from industrial and mixed domestic and industrial waste waters. The nitrifying activity of the sludge is verified by testing in the presence and absence of a specific inhibitor. If the nitrification rate is within a suitable range for the test, i.e. 2 mg of nitrogen per gram of suspended solid and hour to 6,5 mg of nitrogen per gram of suspended solids and hour, the sludge may be used directly. If not, adjustments are necessary. The method is applicable to water-soluble, non-volatile chemicals, and to waste waters
Abstract
Overview
EN ISO 9509:2006 - "Water quality - Toxicity test for assessing the inhibition of nitrification of activated sludge microorganisms" - specifies a laboratory method to assess short‑term inhibition of nitrifying bacteria in activated sludge. The test estimates inhibition over an exposure period typically of 3 hours (extendable up to 24 hours for weakly nitrifying sludge) and is applicable to water‑soluble, non‑volatile chemicals and to whole wastewater samples. The standard is the CEN-adopted version of ISO 9509:2006 and targets water quality and ecotoxicity testing of wastewater treatment processes.
Key Topics and Requirements
- Objective: Measure reduction in production of oxidized nitrogen (nitrite + nitrate) or decreased ammonium removal caused by a test substance or sample.
- Test material scope: Pure chemicals, defined mixtures, chemical products, wastewater and treated wastewater; suitable for sludges from domestic, industrial or mixed sources.
- Exposure time: Normally 3 h; up to 24 h for weakly nitrifying sludge.
- Sludge activity verification: Use a specific inhibitor (e.g., N‑allylthiourea, ATU) to verify nitrifying activity. Sludge is acceptable if nitrification rate lies between 2.0 and 6.5 mg N · g SS⁻¹ · h⁻¹; otherwise adjustments are required.
- Measured endpoints: Oxidized nitrogen (nitrite + nitrate) produced or ammonium consumed under standard conditions after aeration.
- Test logistics: Includes reagents, culture media (full medium and media for wastewater samples), apparatus, preparation of stock solutions, pH adjustment (standard pH ≈ 7.6 ± 0.1), sample storage guidance and calculation/validation procedures.
- Safety: Standard warns of pathogenic organisms in sewage and advises normal laboratory biosafety and chemical handling precautions.
- Documentation: Test report elements, validity checks and precision statements are specified; annexes provide sludge activity determination, example preparations and culturing guidance.
Applications and Users
- Environmental and water quality laboratories performing nitrification inhibition tests.
- Wastewater treatment plant (WWTP) operators monitoring process sensitivity to industrial discharges.
- Regulatory agencies assessing effluent toxicity and compliance.
- Chemical manufacturers and formulators evaluating potential inhibitory effects of new products on biological treatment.
- Consultants and researchers studying impacts of contaminants on activated sludge nitrification.
