ISO 14698:2003 establishes the principles and basic methodology of a formal system of biocontamination control (Formal System) for assessing and controlling biocontamination when cleanroom technology is applied for that purpose. It specifies the methods required for monitoring risk zones in a consistent way and for applying control measures appropriate to the degree of risk involved. In zones where risk is low, it can be used for information.
Abstract
Overview
ISO 14698-1:2003 - Cleanrooms and associated controlled environments - Biocontamination control - Part 1: General principles and methods - defines a structured, risk-based Formal System for assessing and controlling biocontamination in cleanrooms and controlled environments. The standard establishes principles, terminology and basic methodology for consistent monitoring, interpretation and control of viable (microbial) contamination in air, on surfaces, textiles and liquids. It is intended to support hygienic practice where cleanroom technology is applied to protect products, processes and people.
Key topics and requirements
- Formal System: Establish, implement and document a risk-based biocontamination control program integrated with quality management and training.
- Risk zones: Define and classify areas (as-built, at-rest, operational) and designate risk zones to determine monitoring and control measures.
- Alert, action and target levels: Set and review microbiological thresholds (alert and action levels or target levels) based on baseline data and technology limits.
- Monitoring and sampling plan: Use appropriate sampling methods and devices (impact/impingement samplers, settle plates, contact plates, swabs) and a documented sampling plan to detect viable particles without contaminating the product or zone.
- Control measures & corrective actions: Define limits, corrective actions, and verification procedures to respond when levels exceed set thresholds.
- Validation & verification: Provide objective evidence that monitoring devices, laundering processes and procedures are fit for intended use (see annexes for guidance).
- Documentation & training: Maintain documented procedures, training programs and records to ensure consistent biocontamination control and verification.
Important annex guidance (informative):
- Annex A–F: airborne, air sampler validation, surface sampling, textiles, laundering validation, liquid sampling
- Annex G: training guidance
Applications and who uses it
ISO 14698-1 is used by organizations that require microbial control in controlled environments, such as:
- Pharmaceutical, biotechnology and medical device manufacturers
- Healthcare facilities and sterile compounding pharmacies
- Food, cosmetics and other hygiene-sensitive product manufacturers
- Cleanroom designers, validation and quality engineers
- Microbiologists and compliance/regulatory teams
The standard helps these users design monitoring programs, set action/alert levels, select sampling methods, validate processes and demonstrate ongoing control of biocontamination.
