ISO 16000-4:2011 specifies a determination of formaldehyde in indoor air using a diffusive sampler with solvent desorption and high performance liquid chromatography (HPLC). The test method is applicable to the measurement of formaldehyde in indoor air over the range from 0,001 mg/m3 to 1,0 mg/m3 for a sampling period of between 24 h and 72 h. For sampling periods of 24 h, the applicable concentration range is 0,003 mg/m3 to 1 mg/m3, and for 72 h it is 0,001 mg/m3 to 0,33 mg/m3. The method is suitable for measurements in atmospheres with conventional indoor air relative humidity and for monitoring at air velocities as low as 0,02 m/s. The chromatographic step in the method is designed to eliminate potential interferences, including those due to the presence of other carbonyl compounds. The sampling method gives a time-weighted average result.
Abstract
Overview
ISO 16000-4:2011 specifies a validated laboratory procedure for the determination of formaldehyde in indoor air using a diffusive sampling (passive) method with solvent desorption and HPLC analysis. The method yields a time-weighted average concentration and is applicable to typical indoor environments (residences, offices, schools) and controlled test-chamber measurements. It covers measurement ranges from 0.001 mg/m³ to 1.0 mg/m³ depending on sampling duration and is designed to reduce interferences from other carbonyl compounds.
Key Topics
- Sampling principle: Passive/diffusive sampler calibrated by a known diffusive uptake rate; formaldehyde diffuses onto a DNPH-coated silica gel/cellulose strip and forms a stable hydrazone.
- Analytical technique: Solvent desorption (acetonitrile) of DNPH-formaldehyde derivative followed by HPLC-UV analysis for quantification.
- Applicable ranges & sampling periods:
- Overall: 0.001–1.0 mg/m³ for 24–72 h sampling.
- 24 h sampling: 0.003–1.0 mg/m³.
- 72 h sampling: 0.001–0.33 mg/m³.
- Shorter sampling (<24 h) possible if samplers are confirmed for sensitivity and precision.
- Environmental limits: Suitable for conventional indoor relative humidity and air velocities as low as 0.02 m/s.
- Interference control: Chromatographic separation is specified to eliminate potential interferences, including other carbonyl compounds.
- Reagents & safety: Uses 2,4-dinitrophenylhydrazine (DNPH) chemistry. DNPH is hazardous (toxic and explosive when dry) - safety procedures and laboratory best practices are required.
- Quality elements: Calibration standards, desorption efficiency, precision/uncertainty analysis, storage and blank limits, and QA/QC procedures are included.
Applications
ISO 16000-4:2011 is used by:
- Environmental and indoor air quality consultants performing formaldehyde monitoring in buildings.
- Occupational hygienists assessing worker exposure in offices, schools, or industrial settings.
- Research laboratories and academicians studying emissions from building materials (e.g., particleboard, insulation) and indoor exposure.
- Manufacturers and test labs performing emission testing or product certification when passive monitoring is preferred.
- Regulatory bodies and building assessors for compliance checks or long-term surveillance.
Practical advantages include simple deployment (diffusive samplers or personal badges), time-weighted average exposure assessment over 24–72 hours, and reliable HPLC-based specificity for formaldehyde.
