Deals with sampling and methods of detremination. A table shows a summary of the principal characteristics of methods of determination, as referance method, carbon dioxide method, ninhydrin method, flotation method, X-ray method, acoustic method.
Abstract
Overview
ISO 6639-1:1986 - "Cereals and pulses - Determination of hidden insect infestation - Part 1: General principles" sets out the fundamental principles for detecting and measuring hidden insect infestation in cereals and pulses. Part 1 defines terms, explains distribution and behaviour of grain pests, and describes the role of both reference methods and rapid methods for infestation determination. It forms the first element of a multi-part standard (Parts 2–4 cover sampling, the reference method and rapid methods).
Key topics and requirements
- Definitions: Clear distinction between initial observed infestation (free-living insects visible on first inspection) and hidden infestation (insects living inside individual grains, often juvenile stages or those entering via cracks).
- General principles: Insect distribution in bulk grain is non-random and influenced by temperature, moisture and crowding; detection requires specialist sampling and laboratory techniques.
- Sampling considerations: Standard sampling procedures (ISO 950 and ISO 951) are not appropriate for hidden infestation because insects concentrate in specific areas; increments may need to be kept separate if information on distribution is required.
- Reference method: Described as incubation at a standard temperature and relative humidity with periodic examination - a reliable method for developing true-grain insect detection but time-consuming (may require several weeks).
- Rapid methods: Alternatives to shorten detection time. The standard lists and compares methods such as carbon dioxide production, ninhydrin, flotation, X‑ray, and acoustic methods. A summary table in the standard compares principal characteristics (accuracy, suitability for field or lab use, and sensitivity).
- Practical cautions: Handling (e.g., sieving) can kill insects and affect counts; choice of method depends on time, cost and whether results should reflect insect numbers or amount of damaged grain.
Applications and users
ISO 6639-1 is intended for stakeholders concerned with pest detection and quality control of stored cereals and pulses:
- Grain quality laboratories and inspection services
- Food processors, millers and pulse handlers
- Commodity traders and grain storage managers
- Regulatory bodies conducting phytosanitary or quality assessments
Practical uses include selecting appropriate detection methods (reference vs rapid), designing sampling plans for hidden infestation, and interpreting results for trade, storage management and pest-control decisions.
