Abstract
Overview
ISO 6639-4:1987 is Part 4 of the ISO series on “Cereals and pulses - Determination of hidden insect infestation.” This part specifies rapid methods for detecting or estimating hidden insect infestation in cereals and pulses on a qualitative basis. It defines five rapid approaches (carbon dioxide production, ninhydrin, whole‑grain flotation, acoustic, and X‑ray) and gives practical procedures, equipment needs and interpretation guidance for routine grain quality and storage‑pest screening.
Key Topics and requirements
- Scope and applicability
- Rapid qualitative detection of hidden infestation in cereals and pulses (wheat, sorghum, rice, small maize, etc.).
- Large grains (e.g., maize) must be partly broken (kibbled) before testing; this can affect counts of eggs/young larvae.
- Method‑specific limitations (e.g., CO2 method not suitable for finely ground products or products with moisture >15%).
- Rapid methods covered
- Carbon dioxide production (gasometric and infra‑red analyses): incubate a known test portion at 25 ± 1 °C (typically 24 h), measure CO2 generation as a proxy for insect metabolism. Key apparatus: airtight containers (≤750 ml), syringes, incubator, infra‑red analyser or gasometric setup. Typical sample fill ≈300 g; analyser flows described (purified air at ~1 L/min during measurement; some analysers specified to operate with dry air at ≈2000 ml/min).
- Ninhydrin method: crush test portion (free‑insects removed) against ninhydrin‑impregnated paper; insect amino acids develop purple spots. Test portions typically ≥1 000 grains; large grains should be kibbled and re‑sieved.
- Whole grain flotation, acoustic, and X‑ray methods: brief descriptions and intended detection stages. Acoustic detects living feeding larvae/adults; X‑ray can show living and dead larvae/adults; flotation provides qualitative detection.
- Performance and interpretation
- Repeatability criteria given (e.g., gasometric difference ≤0.2% vol; infra‑red difference ≤2 µl·L−1·min−1).
- Thresholds for interpreting CO2/IR results (examples: IR <1.0 µl·L−1·min - probably no infestation; IR ≥6.0 - heavy infestation). Guidance on follow‑up actions (repeat test, fumigation, unsuitable for storage).
Applications and users
- Who uses it: grain elevators, storage operators, quality control laboratories, cereal processors, grain inspectors, fumigation services and agricultural research labs.
- Practical uses:
- Rapid screening of bulk samples to decide whether further detailed testing or remedial action (fumigation) is needed.
- Routine monitoring of stored cereal and pulse lots to protect quality and marketability.
- Supporting sampling and analytical workflows defined in other ISO grain standards.
